We need more information.
What kind of sound card?
What is the error you get?
The more information you give the better. There are a number of reason why you have no sound. Make sure the correct audio drivers for your card are installed. Just updating Windows dosn't guarantee that the driver was part of the updates.

Check you have installed the sound card correctly if you have one. If you are using onboard sound, have you put the mobos cd in and installed everything off there again? I reinstallation of windows will need all component's cd inserted and installed

All Good advice above,
I would put my money on it being a driver problem, if you can tell us what Mobo, or sound card you have, or failing that what make and model your machine is, we can find the drivers for you and give you a walk through on what to do, that is if you need it.
Don't be scared to ask if you don't know something or want more info about something :)
